What is Supprelin LA?

Supprelin LA is an FDA (U.S. Food and Drug Administration) approved medication manufactured by Endo Pharmaceuticals Solutions Inc. Supprelin La (histrelin) is classed as a gonadotropin releasing hormone (GnRH) agonist and is used for the treatment of children with central precocious puberty (CPP).

CPP causes puberty to come early in girls (before 8 years of age) and in boys (before 9 years of age). Signs of early puberty include breast enlargement in girls and the appearance of hair in the genital area in boys and girls. CPP can have long-lasting effects. These effects include increased bone age. Histrelin works by reducing the amount of testosterone in boys and estrogen in girls.

Vantas, another type of histrelin implant is used to treat the symptoms associated with advanced prostate cancer.

What doses of Supprelin LA are available?

Supprelin LA is available as an implant, in the following doses: 50 mg subcutaneous implant which delivers approximately 65 mcg histrelin acetate per day over 12 months.

Use an anesthetic to numb the area. The implant is inserted subcutaneously in the inner aspect of the upper arm. Use absorbable sutures and/or sterile adhesive surgical strips to close the incision. Keep the bandage in place for several days until the surgical incision heals. Avoid any heavy lifting, bumping of the incision site, or physical activity for 7 days after the procedure.

Supprelin LA active ingredients

The active ingredient histrelin acetate results in an increase in circulating levels of LH and FSH, leading to a transient increase in the concentration of gonadal steroids (testosterone and dihydrotestosterone in males, and estrone and estradiol in premenopausal females).

Continuous use of histrelin acetate causes a reversible down-regulation of the GnRH receptors in the pituitary gland and desensitization of the pituitary gonadotropes resulting in decreased levels of LH and FSH.

Supprelin LA side effects

The most common side effects of Supprelin LA in clinical trials include:

  • Implant site reaction, including complications related to implant insertion or removal – bruising, pain, soreness, erythema, and swelling
  • Suppression of endogenous sex steroid secretion may occur
  • Light vaginal bleeding in girls

In rare instances, Supprelin LA can cause more serious side effects. These can include:

  • Initial transient increases of estradiol and/or testosterone may cause a temporary worsening of symptoms
  • Increase in tumors of hormonally responsive tissues

Other drugs may interact with Supprelin LA, including prescription drugs and over-the-counter medicines, vitamins, and herbal products. Tell your health care provider about all of your medicines including any supplements you are taking.

Supprelin LA contraindications

You should not use Supprelin LA if you:

  • Are allergic to the active ingredient histrelin or to similar drugs such as leuprolide
  • Have had an allergic reaction to any of the other ingredients in Supprelin LA
  • Have a history of hypersensitivity to gonadotropin-releasing hormone (GnRH) or GnRH analogs
  • Are a pediatric patient less than 2 years of age
  • Are pregnant or are planning to become pregnant

Talk to your doctor before using Supprelin LA if you:

  • Are taking any of the medications that could interact with Supprelin LA
  • Are breastfeeding or are planning to breastfeed
