What is Malarone?

Malarone (atovaquone/proguanil) is an FDA (U.S. Food and Drug Administration) approved medication manufactured by GlaxoSmithKline. It is an antimalarial drug used for the prophylaxis of Plasmodium falciparum malaria infection, and in areas where chloroquine resistance has been identified, and for the treatment of acute, uncomplicated P. falciparum malaria.

What doses of Malarone are available?

Malarone is available in tablet form, in the following doses: 250 mg atovaquone and 100 mg proguanil hydrochloride. It is also available as Malarone Pediatric tablets, in the following doses 62.5 mg atovaquone and 25 mg proguanil hydrochloride. The tablets are round, film-coated, tablet embossed with “GX CM3.

Malarone tablets should be taken with food or a milky drink. Start prophylactic treatment 1 or 2 days before entering a malaria area. Continue daily for the duration of the stay and for 7 days after return. Adults will take one tablet daily, pediatric patients’ dosage is based on body weight.

No medication is 100% effective in the treatment or prevention of malaria. Use protective clothing, insect repellents, and mosquito netting to also prevent mosquito bites that could cause malaria.

Do not use Malarone for the prophylaxis of malaria in patients with severe renal impairment and use it with caution for the treatment of malaria in patients with severe renal impairment. Malarone active ingredients

Malarone contains atovaquone and proguanil hydrochloride as the active ingredients. Atovaquone is classified as a selective inhibitor of parasite mitochondrial electron transport.

Malarone side effects

The most common side effects of Malarone in clinical trials include:

  • Diarrhea
  • Oral ulcers
  • Headache, dizziness
  • Weakness
  • Cough
  • Abdominal pain
  • Nausea, vomiting, loss of appetite, anorexia
  • Pruritus

In rare instances, Malarone can cause more serious side effects. These can include:

  • Neutropenia, anemia, pancytopenia
  • Elevated liver laboratory tests and cases of hepatitis and hepatic failure requiring liver transplantation

Malarone drug interactions

Malarone can interact with other medications. These include:

  • Rifampin or rifabutin
  • Warfarin and other coumarin-based anticoagulants
  • Tetracycline may reduce atovaquone concentrations
  • Metoclopramide

Malarone contraindications

You should not use Malarone if you:

  • Are allergic to the active ingredients atovaquone or proguanil hydrochloride
  • Have had an allergic reaction to any of the other ingredients in Malarone

Talk to your doctor before using Malarone if you:

  • Are taking any of the medications that could interact with Malarone
  • Are pregnant or are planning to become pregnant
  • Are breastfeeding or are planning to breastfeed

Malarone is the trademark brand name for atovaquone and proguanil hydrochloride manufactured by GlaxoSmithKline. A generic version of atovaquone and proguanil hydrochloride is available.
